In this work, I explore the gut microbiome as a central factor in the development and progression of autoimmune diseases. Rather than viewing autoimmunity solely through the lens of genetics and classical immunology, this monograph emphasizes the dynamic interaction between host and microbiota as a key determinant of immune tolerance and systemic inflammation. The study examines the molecular mechanisms underlying microbiome-immune crosstalk, including regulatory T-cell modulation, Th17 differentiation, short-chain fatty acid signaling, intestinal barrier integrity, molecular mimicry, metabolic regulation, and epigenetic influence. By integrating current scientific evidence from experimental models, clinical studies, longitudinal research, and metagenomic analyses, this work highlights how microbial imbalance contributes to immune dysregulation.Special attention is given to emerging diagnostic and therapeutic perspectives. Microbiome-based biomarkers, dietary strategies, probiotics, prebiotics, fecal microbiota transplantation, and personalized approaches are discussed as promising directions in modern precision medicine.
